Extract of Aquilaria crassna leaves and mangiferin are vasodilators while showing no cytotoxicity

Abstract: The leaves of Aquilaria spp. promote “physiological balance”, and are “cardiotonic and provide blood nourishment”. In Asia, these leaves are increasingly consumed as tea and claimed to provide benefits to cardiovascular function, albeit without any scientific proof. Therefore, this study sought to evaluate the action of Aquilaria crassna leaf aqueous extract (AE) on vascular function and vascular smooth muscle cytotoxicity. AE and a main constituent, mangiferin wer… Show more

“…Mangiferin's safety as well as its beneficial effect in improving cellular function was reported by several studies (Mei et al 2021b). The vascular protection effect of mangiferin in a range from 0.1 to 100 μM was proven without any cytotoxicity (Wisutthathum et al 2019). Also bone marrow erythrocytes showed no genotoxicity after oral application of 2 g/kg of mangiferin to NMRI mice (Rodeiro et al 2012).…”

“…For cytotoxicity testing by the MTT assay, 26 cells were incubated with MTT reagent (0.5 mg/mL) for 4 h at 37 °C in CO 2 incubator. Then, the solution was discarded and DMSO (100 μL) was added and shaken for 15 min to dissolve formazan crystals.…”

“…In some experiments, endothelial cells were mechanically removed by gently rubbing the lumen with a stainless steel wire. The mesenteric rings were mounted on a pair of intraluminal wires in organ chambers containing physiological Krebs’ solution (mM): NaCl, 122; KCl, 5; [N-(2-hydroxyethyl) piperazine N’-(2-ethanesulfonic acid)] HEPES, 10; KH 2 PO 4 , 0.5; NaH 2 PO 4 , 0.5; MgCl 2 , 1; glucose, 11; and CaCl 2 , 1.8 (pH 7.3), at 37 °C and aerated [29,49,50,51]. The vessel segments were allowed to equilibrate for 1-h at a resting tension of 1–1.3 g during which the solution was replaced every 15 min.…”
