Extensive localization of long noncoding RNAs to the cytosol and mono- and polyribosomal complexes

Abstract: BackgroundLong noncoding RNAs (lncRNAs) form an abundant class of transcripts, but the function of the majority of them remains elusive. While it has been shown that some lncRNAs are bound by ribosomes, it has also been convincingly demonstrated that these transcripts do not code for proteins. To obtain a comprehensive understanding of the extent to which lncRNAs bind ribosomes, we performed systematic RNA sequencing on ribosome-associated RNA pools obtained through ribosomal fractionation and compared the RNA… Show more

“…84 Molecular mechanisms by which SPRY4-IT1 affects melanoma progression require further investigation. RNA-FISH analysis showed that this lncRNA is predominantly localized in the cytoplasm of melanoma cells 67 , and an association with polysomes has been demonstrated. 85 SPRY4-IT1 associates with, and reduces the abundance of, the lipid phosphatase lipin 2 and may suppress apoptosis arising from lipid metabolism and lipotoxicity.…”

“…Perhaps a significant fraction of mRNA-like long intervening non-protein-coding RNAs are mRNAs after all, encoding peptides and short proteins. The question on how many of the non-protein-coding RNAs are associated with polysomes for productive translation is currently under debate (Ingolia et al 2011;Guttman et al 2013;Ingolia et al 2013;Ulitsky and Bartel 2013;van Heesch et al 2014). …”

“…Thousands of lncRNAs have been annotated in eukaryotic genomes (Derrien et al 2012;Ulitsky and Bartel 2013), many of which are preferentially located in the cytoplasm (van Heesch et al 2014), where they can engage in miRNA-mediated interactions with other transcripts. Both computational and experimental evidence support the extensive targeting of lncRNAs by miRNAs (Paraskevopoulou et al 2013).…”
