Expression studies of neuronatin in prenatal and postnatal rat pituitary

Abstract: The pituitary gland, an indispensable endocrine organ that synthesizes and secretes pituitary hormones, develops with the support of many factors. Among them, neuronatin (NNAT), which was discovered in the neonatal mouse brain as a factor involved in neural development, has subsequently been revealed to be coded by an abundantly expressing gene in the pituitary gland but its role remains elusive. We analyze the expression profile of Nnat and the localization of its product during rat pituitary development. The… Show more

“…6 However, the role of NNAT in vivo is poorly understood. Recently, we found that NNAT first appears in and occupies stem/progenitor cells in the rat embryonic pituitary primordium and is then downregulated during differentiation into mature hormone-producing cells, suggesting that NNAT may be involved in pituitary development 7 Moreover, we found that NNAT localizes in various subcellular organelles in addition to the endoplasmic reticulum (ER), as reported previously. 8…”

“…In the pancreas, NNAT localizes in the ER of β-cells, 8 However, in a previous study of the pituitary gland, we observed, for the first time, that NNAT was widely localized in subcellular organelles in addition to the ER. 7 Consequently, we have re-examined subcellular localization of NNAT in the adult pancreas using an antibody against PDI, a marker of the ER (Fig. 3A).…”

“…For example, the NNAT product, neuronatin, is expressed in prenatal and postnatal rat pituitary [23]. However, the role of neuronatin in the adult pituitary has not been clarified yet.…”

“…Other markers expressed in the SOX2 stem cell niche include Sox9, Oct4, Gfra2, Ret, Ly6a (Sca-1), Prop1, Prrx1, Cdh1 (E-cadherin), S100β , β-catenin, neuronatin, neurturin, and Cxadr (Coxsackievirus and adenovirus receptor) (3,73,75,76,7981). Clonogenic potential has been demonstrated for cells expressing Sox2, Sox9, Gfra2 , and S100b , but the remaining markers have not been tested yet.…”
