Expression, purification, crystallization and preliminary X-ray analysis of perakine reductase, a new member of the aldo-keto reductase enzyme superfamily from higher plants

Abstract: Perakine reductase (PR) is a novel member of the aldo-keto reductase enzyme superfamily from higher plants. PR from the plant Rauvolfia serpentina is involved in the biosynthesis of monoterpenoid indole alkaloids by performing NADPH-dependent reduction of perakine, yielding raucaffrinoline. However, PR can also reduce cinnamic aldehyde and some of its derivatives. After heterologous expression of a triple mutant of PR in Escherichia coli, crystals of the purified and methylated enzyme were obtained by the hang… Show more

“…Several refined [STR (Ma et al, 2006) and SGD ] and partial [raucaffricine glucosidase , perakine reductase (Rosenthal et al, 2006) and vinorine synthase ] crystal structures for enzymes involved in MIA biosynthesis in R. serpentina have recently been reported (Figure 2). The information obtained from the crystal structure of STR complexed with strictosidine was used to produce the rational site-directed V208A mutation of STR.…”

“…The absence of both a Rossmann fold (11) and the catalytic motif Tyr-Xaa-Xaa-Xaa-Lys (12) (13). Furthermore, sequence alignment and preliminary x-ray analysis of a methylated PR mutant support the view that PR represents a novel member of the AKR superfamily (6,14). In fact, PR would be the first example of a plant AKR involved in indole alkaloid biosynthesis in contrast to the plant AKR-type codeinone reductase, which is involved in isoquinoline alkaloid metabolism (15).…”

“…The availability of crystal structures for various enzymes involved in MIA biosynthesis, namely STR, SGD, raucaffricine glucosidase, perakine reductase and vinorine synthase (Ma et al, , 2006Rosenthal et al, 2006;Ruppert et al, 2006;Barleben et al, 2007), have set the stage for rational protein engineering and for altering the substrate specificity of these reactions. In the case of STR, crystal structures were produced with the enzyme complexed to secologanin or to tryptamine.…”
