Expression Profiling of Mammalian Male Meiosis and Gametogenesis Identifies Novel Candidate Genes for Roles in the Regulation of Fertility

Abstract: We report a comprehensive large-scale expression profiling analysis of mammalian male germ cells undergoing mitotic growth, meiosis, and gametogenesis by using high-density oligonucleotide microarrays and highly enriched cell populations. Among 11,955 rat loci investigated, 1268 were identified as differentially transcribed in germ cells at subsequent developmental stages compared with total testis, somatic Sertoli cells as well as brain and skeletal muscle controls. The loci were organized into four expressio… Show more

“…It is unique among integrated databases because it is focused on specific biological components and processes that play roles in germ-cell biology. It also provides access to microarray and image data relevant to the knowledge base [Schlecht et al, 2004;Zhou et al, 2005;Hermida et al, 2006]. Future goals for GermOnline include adding capacity to curate data on protein-DNA interactions and SNPs, and adding video and other types of image data.…”

“…1,2 In the last 25 years, advances in molecular biology and genomics have significantly improved our knowledge of spermatogenesis by identifying numerous genes essential for the process. [3][4][5][6] However, these transcriptomic databases do not provide crucial information on the post-transcriptional control of gene expression, changes in protein expression levels or protein modifications. Moreover, transcriptomics poses certain limitations as the expression levels of the majority of mRNAs and proteins are not always correlated.…”

“…spermiogenesis), while other cell types are obtained at very low purity levels (18). Consequently, most differential gene expression studies on isolated spermatogenic cell populations are based on the comparison of transcripts specifically identified in P spermatocytes and RS (e.g., 24,33,34).…”
