Expression profiles of exosomal tRNA-derived fragments and their biological functions in lipomas

Abstract: There is evidence that exosomes derived from the lipoma tissue (Exo-LT) have a stronger capacity to promote the proliferation and migration of adipose-derived stem cells (ADSCs) than those from the adipose tissue (Exo-AT). But the Exo-LT do not have a significant effect on the adipogenic differentiation of the ADSCs. Recently, certain exosomal tRNA-derived fragments (tRFs) have been shown to play a crucial role in the pathogenesis of certain tumors. Therefore, it is necessary to identify the differently expres… Show more

“…Our study is the rst giving comparative analysis of Exo-LT and Exo-AT at the lncRNA, miRNA, and mRNA levels. In our previous study, we have analyzed difference in tRNA-derived fragments (tRFs) expression from Exo-LT and Exo-AT, and by identifying the differentially expressed tRFs in Exo-LT, we have preliminarily explained the reason for Exo-LT have a stronger capacity in promoting the proliferation and migration of ADSCs than Exo-AT (22,40). Now we have comprehensively analyzed the differences of lncRNAs, miRNAs and mRNAs expression in Exo-LT and Exo-AT, and constructed three ceRNA networks in Exo-LT, which we suggest that the unique expression of genes in Exo-LT affects its roles in promoting the proliferation and adipogenesis of ADSCs.…”

“…Currently, with the application of high-throughput sequencing and the advances in bioinformatics techniques, increasing number of tRFs in extracellular vesicles have been reported to be associated with different disease [21] .It has been reported that 5′-tRFs wer present at different levels between preeclamptic and normotensive trophoblast tissues from the placenta [22] .Zhu et al demonstrated the presence of tRFs in exosomes of liver cancer cells [23] .Koi et al con rmed that the expression levels of tRF-Lys-TTT (tRF-32-PS5P4PW3FJHP1, 5′-tRNA half) were signi cantly elevated in breast cancer [24] .Zhang et al identi ed 11 upregulated tRFs and 18 downregulated tRFs in osteoporosis compared with healthy controls, and the expression levels of tRF-25-R9ODMJ6B26 (3′-tRF), tRF-38-QB1MK8YUBS68BFD2 (3′-tRF), and tRF-18-BS68BFD2 (3′-tRF) were signi cantly higher in osteoporosis [25] .…”
