Expression patterns of signaling lymphocytic activation molecule family members in peripheral blood mononuclear cell subsets in patients with systemic lupus erythematosus

Abstract: Genome-wide linkage analysis studies (GWAS) studies in systemic lupus erythematosus (SLE) identified the 1q23 region on human chromosome 1, containing the Signaling Lymphocytic Activation Molecule Family (SLAMF) cluster of genes, as a lupus susceptibility locus. The SLAMF molecules (SLAMF1-7) are immunoregulatory receptors expressed predominantly on hematopoietic cells. Activation of cells of the adaptive immune system is aberrant in SLE and dysregulated expression of certain SLAMF molecules has been reported.… Show more

“…Costimulatory molecule and microbial sensor SLAMF1 (also known as CD150) initiates signal transduction pathways in a variety of immune cells [ 24 ]. SLAMF1 is overexpressed on the T cell, B cell and their subgroups of Systemic lupus erythematosus (SLE) patients [ 25 , 26 ]. Expression of SLAMF1 was significantly increased in peripheral blood CD4+, T helper 17, and CD19+ B cells from autoimmune thyroid disease patients [ 27 ].…”

“…B cells are potent antigen presenting cells, able to initiate and maintain T cell responses, both through cell-cell contact and cytokine release. SLAMF1 is upregulated on both T cells and B cells of patients with SLE (12). We examined the effect of SLAMF1 ligation using a specific anti-human SLAMF1 mAb in the context of T-B interactions in vitro .…”

“…The −262 A/G and −188 A/G polymorphisms in the promoter region of SLAMF1 are linked to higher SLAMF1 mRNA expression following stimulation of SLE peripheral blood mononuclear cells with phytohemagglutinin and may contribute to increased SLE susceptibility (10). Moreover, SLAMF1 is up-regulated on the cell surface of both T cells and B cells in the peripheral blood of patients with SLE, suggesting a potential role of SLAMF1 in SLE immunopathogenesis (11, 12).…”

“…Several studies have examined the expression of SLAMF1 to 7 receptors on the surface of T cells isolated from the peripheral blood of SLE patients in comparison to healthy controls, and delivered variable results, thus reflecting the heterogeneity of the disease. 120127 Two studies systematically examined the expression of SLAMF1 to 7 receptors on the T cells from cohorts of patients suffering from SLE. 120,121 In one, authors examined the expression of SLAMF receptors on T cells isolated from patients with lupus nephritis and showed that expression of SLAMF3, SLAMF5, and SLAMF7, on CD8 + and DN T cells was lower in patients who are in remission as compared to patients with active nephritis.…”
