Expression of Versican and ADAMTS During Rat Tooth Eruption

Abstract: A disintegrin and metalloprotease with thrombospondin type 1 motifs (ADAMTS) is a family of extracellular proteases and implicated in cleaving proteoglycans, such as aggrecan, versican and brevican. No information is available about expression or localization of these ADAMTSs in teeth. Versican is a large chondroitin sulfate proteoglycan that is present in a variety of connective tissue including dental pulp, dentin, cementum and periodontal ligaments. The present study was designed to investigate expression o… Show more

“…Osteoblasts and osteocytes may play a role in remodeling of the ECM during bone healing by coordinating the production and degradation of ECM proteins. Similar observations have been reported during bone development Tsubota et al, 2002;Maruya et al, 2003;Nakamura et al, 2005;Sone et al, 2005) and during appositional bone formation (Hatori et al, 2004).…”

“…The degradation products of the versican V1 variant were detected by Western blotting analysis in the porcine predentin matrix, and this was confirmed by ADAMTS4 digestion 23) . These results support the immunochemical detection of versican in the predentin as reported in the previous study 2) .…”

“…Several proteolytic activities are present in the predentin and dentin 1,19) , including kallikrein 4 (Klk4), matrix metalloproteinase 2(Mmp-2), Mmp-9, a disintegrin and metalloproteinase with thrombospondin motifs 4 (Adamts4), and Mmp-20 [20][21][22][23][24] . These activated proteinases degrade the coexisting proteoglycans following the addition of calcium in vitro 2) .…”

“…Dental pulp cells, odontoblasts, cementoblasts, cementocytes, osteoblasts, osteocytes, and periodontal ligament cells use ADAMTS5, which is one of the lytic enzymes that regulate the extracellular accumulation of proteoglycans, and ADAMTS5 plays a key role in remodeling the extracellular environment surrounding these cells (11). It has also been postulated that remodeling of proteoglycans may contribute to tooth eruption (12,13).…”
