Expression of VEGFR-2 during Liver Regeneration after Partial Hepatectomy in a Bioluminescence Mouse Model

Abstract: Background: Liver regeneration requires the formation of new blood vessels. Endothelial cell proliferation is stimulated by vascular endothelial growth factor (VEGF) and its receptor tyrosine kinase VEGFR-2. The aim of this study was to investigate VEGFR-2 expression in vivo during liver regeneration after partial hepatectomy (PHx). Methods: Transgenic VEGFR-2-luc mice were used in which the luciferase reporter gene was under control of the VEGFR-2 promoter. Following 2/3 PHx, the mice underwent in vivo biolum… Show more

“…Therefore, we analyzed the angiogenic endothelial cell activity and microvessel density in the liver after PHx by immunohistochemistry. A markedly increased mean angiogenic activity was detected at day 4 after PHx that decreased steadily until day 21 which is in accordance with previously published data (Alizai et al, 2017). Quantitative analysis of the microvessel density showed no significant differences between mice after PHx and control mice.…”

“…The monitoring of ARE activity in liver regeneration post partial hepatectomy (PHx) was made possible in vivo by using an ARE-driven luciferase reporter gene from the firefly ( Photinus pyralis ) [210] . Nrf2 activity was measured through in vivo bioluminescence imaging and in vitro luminescence assays.…”

“…VEGFR-1 signaling facilitated liver recovery by reconstitution of sinusoids through recruitment of VEGFR-1expressing macrophages and by affecting gene expression including hepatotrophic and pro-angiogenic growth factors in mice [88]. Furthermore, VEGFR-2 activity showed a significant increase after partial hepatectomy in transgenic VEGFR-2-luc mice with maximum signals recorded on POD 3 [89]. However, data on humans are sparse.…”
