Expression of tissue inhibitor of metalloproteinases-1 and B-cell lymphoma-2 in the synovial membrane in patients with knee osteoarthritis

Abstract: The purpose of this study was to determine the expression and impact of tissue inhibitor of metalloproteinases-1 (TIMP-1) and B-cell lymphoma-2 (Bcl-2) in knee osteoarthritis (KOA). We collected synovial fluids from the knee joint of 70 KOA patients and 30 controls. The expression levels of TIMP-1 and Bcl-2 were significantly higher in KOA patients than those in the control group (P<0.01). We also found positive correlation between the severity of KOA and the expression level of TIMP-1 (r=0.8027, P<0.05) and a… Show more

“…The relative expression of TIMP-1 and B-cell lymphoma/ leukemia-2, an anti-apoptotic protein that promotes cell survival by inhibiting apoptosis, were significantly increased in the SF of KOA patients compared with the control group 42 . Moreover, the expression levels of both markers in the synovial tissues of KOA patients were positively correlated with KOA severity.…”

“…42 TIMP-1 is an inhibitor of MMPs, and therefore considered a marker of anabolic activity, increased levels have been demonstrated in OA. 48 A significant increase was shown at 3 weeks following KJD 42 and 6 weeks following ACI 43 with no significant increase seen following HA injection. 40 TGFβ-1 is a growth factor involved in cartilage maintenance.…”

“…46 IL-8 is a chemoattractant cytokine and has been previously associated with WOMAC scores in knee OA. 47 TIMP-1 is an inhibitor of MMPs with increased levels seen in OA patients compared to controls 48 and may be predictive of progression of radiographic changes in hip OA. 49 The other biomarkers investigated are described further in the discussion.…”

“…Excessive synovial fluid stimulates inflammatory cells to produce the inflammatory factors, causing pain and swelling in the joints ( McInnes and Schett, 2011 , Szekanecz et al, 1994 ). Meanwhile, inflammatory cells secrete many cytokines, such as matrix metalloproteinases (MMPs) and adhesion molecules, which play important role in the destruction of bone, further leading to loss of function ( Lee et al, 2012 , Li et al, 2015 , Li et al, 2018 ). Hence, anti-inflammatory therapy is always the preferred treatment option for RA patients in clinic ( Ten Klooster, de Graaf, & Vonkeman, 2019 ).…”
