Expression of the SARS-CoV-2 Receptor ACE2 in Human Retina and Diabetes—Implications for Retinopathy

Abstract: PURPOSE.To investigate the expression of angiotensin-converting enzyme 2 (ACE2), the receptor for SARS-CoV-2 in human retina. METHODS.Human post-mortem eyes from 13 non-diabetic control cases and 11 diabetic retinopathy cases were analyzed for the expression of ACE2. To compare the vascular ACE2 expression between different organs that involve in diabetes, the expression of ACE2 was investigated in renal specimens from nondiabetic and diabetic nephropathy patients. Expression of TMPRSS2, a cell-surface proteas… Show more

“…Interestingly, in the eye, ACE-2 receptors are expressed in the retinal ganglion cell layer, inner plexiform layer, inner nuclear layer, and photoreceptor outer segments. On the other hand, TMPRSS2 is also expressed in multiple retinal neuronal cells, vascular and perivascular cells, as well as in retinal Müller glial cells [ 35 ]. ACE-2 receptor expression was also shown in astrocytes and oligodendrocytes in human and rodent brains [ 36 ] and in microglial cells [ 37 ], suggesting possible viral entry in the retinal cells.…”

“…One of the explanation could be a vascular obstruction caused by a direct viral tissue injury and infiltration of the endothelial cells via the ACE2 receptor. ACE2 receptor are expressed in multiple neuro-retinal cells including the retinal ganglion cell layer, inner plexiform layer inner nuclear layer, and photoreceptor outer segments [ 18 ].…”

“…We performed IHC as previous described ( 53 , 56 ). Deparaffinized sections were boiled in 1× target retrieval solution (Dako; Agilent Technologies) for 20 minutes.…”
