Expression of the mouse interleukin‐2 receptor γ chain in various cell populations of the thymus and spleen

Abstract: Expression of the gamma chain, which is shared among functional receptor complexes for interleukin (IL)-2, IL-4 and IL-7, was examined with hematopoietic cells in mouse thymus and spleen by flow cytometry. The gamma chain was expressed in cell populations from the spleen. Stimulation with concanavalin A and lipopolysaccharide caused fluctuation in expression of the gamma chain in T and B cells, respectively. T lineage cells developing in the adult thymus expressed the gamma chain. Fetal thymus at day 15 contai… Show more

“…Neutralizing anti-c-Kit (ACK2) and neutralizing anti-IL-7R␣ (A7R34) antibodies were kind gifts from S. Nishikawa (Kyoto University, Japan). Because virtually all thymocytes express the common cytokine receptor ␥ chain, thymocytes that express the IL-7R␣ chain should possess a functional IL-7R heterodimer (13). These antibodies were directly conjugated with phycoerythrin (PE), fluorescein-5-isothiocyanate (FITC), allophycocyanin, or Texas Red.…”

“…The ␤ and ␥ chains, which are shared with other interleukin receptors (4,5), belong to the family of hematopoietic cytokine receptors. Resting T cells constitutively express the ␥ chain (6,7) and basal levels of the ␤ chain (8). IL-2R␤ expression increases upon antigen stimulation (9), 2 .…”

“…These findings are consistent with previously published data on the expression of CD122 in mouse lymphoid tissues. 19,20 In fsn/fsn PLN, CD122 was expressed at low levels by a small minority of CD4 + cells. However, the most striking and consistent feature of fsn/fsn PLN was the dramatically increased expression of CD122 on CD8 + cells.…”
