Expression of suppressor of cytokine signaling 3 (SOCS3) and interleukin-6 (-174-G/C) polymorphism in atopic conditions

Abstract: Hypersensitivity of the immune system is caused by elevated immunoglobulin E (IgE) levels in the serum, in response to a discrete allergen leading to allergic reactions. IgE-mediated inflammation is regulated by the cascade of defense related signaling molecules including interleukin-6 (IL-6) that plays pivotal role in the survival and maturation of mast cells during an allergic reaction. IL-6 mediated defense responses are tightly regulated by Suppressor of Cytokine Signaling 3 (SOCS3), an inhibitory molecule… Show more

“…In mouse models of allergic asthma and conjunctivitis, SOCS3 overexpression has been found 60,61 . Elevated expression of SOCS3 was reported in patients with atopic disorders, including food allergy, allergic rhinitis, allergic asthma, atopic dermatitis, and allergic conjunctivitis 56 . Overexpression of SOCS3 was detected in skin lesions in atopic dermatitis patients 62,63 .…”

“…It was proposed that females are more susceptible to atopic illnesses due to the overexpression of SOCS3 in females than males. 56 Infection with helminths has a negative correlation with allergies. Infection with Schistosoma haematobium imparted protection against atopic hypersensitivity through downregulation of SOCS3.…”

“…In immune cells, including MCs, the expression of cytokines is negatively controlled by the activity of the cytokinesuppressor-proteins (SOCS) (18)(19)(20). To understand whether SOCS protein could be involved in the different responses of primed MC to re-stimulation, we investigated the level of mRNA expression of SOCS3 and the methylation profile of SOCS3 promoter by rt-MSP, in BMMCs pre-stimulated with LPS.…”

“…SOCS3 is known to cause an allergic response through its Th2 activity. 11 Single-nucleotide polymorphisms (SNPs) may be present in other gene regions, such as 5′-or 3′untranslated regions (UTRs), introns, or promoters, as well as in the exonic region. Genetic variations in the 3′-UTR can modify gene expression by miRNA binding, protein-mRNA interactions, gene expression disruption, and polyadenylation; thus, SNPs in 3′-UTRs are intriguing for researchers.…”

“…SOCS3 activity is controlled by IL‐6, so change in serum levels of IL‐6 can affect SOCS3 expression and activity. SOCS3 is known to cause an allergic response through its Th2 activity 11 …”
