Expression of sialyl-Tn sugar antigen in bladder cancer cells affects response to Bacillus Calmette Guérin (BCG) and to oxidative damage

Abstract: The sialyl-Tn (sTn) antigen is an O-linked carbohydrate chain aberrantly expressed in bladder cancer (BC), whose biosynthesis is mainly controlled by the sialyltransferase ST6GALNAC1. Treatment with Bacillus Calmette-Guérin (BCG) is the most effective adjuvant immunotherapy for superficial BC but one third of the patients fail to respond. A poorly understood correlation between the expression of sTn and BC patient's response to BCG was previously observed. By analyzing tumor tissues, we showed that patients wi… Show more

“…Worth mentioning, IL-6, which is mainly held responsible for cytokine release syndrome, was detected in supernatants of samples containing target cells regardless of the presence or absence of the αSTn-IgG4 TM. This suggests that IL-6 is released by MDA-MB-231 and MCR cancer cells and not by activated UniCAR T-cells, which is in line with our previous publication and other studies [12,35,36]. As stated before, proinflammatory cytokines are specifically released following TMdependent cross-linkage of tumor and UniCAR T-cells.…”

“…For this reason, we investigated the effect of B4GALNT2 on the transcriptome of transfected LS174T cell variants. The ability to modulate the transcriptome of cancer cells by a glycosyltransferase was previously documented [17,[24][25][26]. Nevertheless, the transcriptomic changes induced by B4GALNT2 in standard conditions of growth were surprisingly relevant.…”

“…Sialyl-Tn-and sialyl-Tn/S6T-expressing bladder tumors were treated with bacillus Calmette-Guerin (BCG) [88,89]. Lower recurrence rates of sialyl-Tn-positive tumor cells were observed post-BCG treatment.…”
