Expression of RNA polymerase IV and V in Oryza sativa

Abstract: RNA polymerase IV and V are principal players in the RdDM pathway, where their current study has shown interaction of several factors that control DNA silencing of intergenic regions and siRNA production. DNA silencing is an important process during cell differentiation, nuclear structure and viral control. However, RNA pol IV and V are yet to be study in model monocot systems like Oryza sativa that can provide further information on genetic silence mechanism in plats. We show the expression pattern of these p… Show more

“…A specific transcriptional response in early development has been described for the genes involved in DNA methylation, which were upregulated during the 6-month follow-up (Jiménez-Ruiz et al, 2018). Interestingly, the role of RNA pols IV and V in RNA-directed DNA methylation has been clearly demonstrated (Huang et al, 2009;Ream et al, 2009;Haag and Pikaard, 2011;Haag et al, 2014;Zhou and Law, 2015) by acting during development (Pikaard and Tucker, 2009;Moo et al, 2012;Haag et al, 2014). However, the NRPE5 gene for RNA pol V (and/or IV) did not specifically and differently modulate its expression.…”

Images were taken using a 40x lens of an Olympus FV100 SW laser confocal microscope, and they were processed using the FV10 ASW software version 4.1.…”

“…Transcription in bacteria and archaea is carried out by a single multimeric RNA polymerase, while most eukaryotes contains three multimeric complexes (RNA pol I, II, and III) [1][2][3]. Furthermore, plants contain two additional RNA pols (IV and V) that have evolved from RNA pol II [4][5][6][7][8][9][10][11][12]. RNA pol I consists of 14 subunits and synthesizes precursor rRNA 45S (35S in yeast) of the three largest rRNAs [3,[13][14][15].…”

“…Plant-specific RNA pol IV and V, which have evolved from RNA pol II through duplication and functional divergence, also contain 12 subunits. These two enzymes, that are involved in epigenetic regulation synthesize siRNAs, which play roles in transcriptional silencing via RNA-directed DNA methylation (RdDM), and non-coding RNAs participating in plant growth, development, response to environmental changes or plant immunity [4][5][6][7][8][9][10][11][12][22][23][24].…”
