Expression of proteoglycan core proteins in human bone marrow stroma

Abstract: Heparan sulphate proteoglycans (HSPGs) present on the surface of bone marrow stromal cells and in the extracellular matrix (ECM) have important roles in the control of adhesion and growth of haemopoietic stem and progenitor cells. The two main groups of proteoglycans which contain heparan sulphate chains are members of the syndecan and glypican families. In this study we have identified the main surface membrane and matrixassociated HSPGs present in normal human bone marrow stroma formed in long-term culture. … Show more

“…HSPGs can be divided into three major classes: (1) lipid-anchored, e.g., the glypicans; (2) transmembrane, e.g., syndecans; and (3) extracellular or secreted, a group that includes perlecan [42]. HSPGs had been found in cartilage [43], bone stroma [44], uterine decidua [45], and follicular fluid [46]. Perlecan is particularly abundant in the ovary with a level of expression that is stronger than the kidney-a known rich source of this protein.…”

“…Therefore, since our data imply a minimal presence of core proteins in Jurkat cells causing poor bacterial attachment, the gene expression of PG-core proteins was assessed using RT-PCR analysis. Syndecan-1, syndecan-2, syndecan-4, betaglycan, glypican-1, and thrombomodulin are expressed in epithelial and endothelial cells [38,39], and CD44 is expressed in lymphocytes [39,40].…”

“…Reportedly, the major PG of lymphocytes is the CD44 molecule, which is expressed on the cell surface and is a component of GAGs such as chondroitin sulfate [38][39][40].…”

“…In the kidney, perlecan is found in the mesangium, Bowman's capsule, the tubular basement membrane, but is only slightly expressed in the glomerular basement membrane (Groffen et al, 1997). In bone, marrow but not the mineral compartment, is rich in perlecan (Schofield et al, 1999). In human fetal tissue (12-14 week old), pericellular perlecan expression was detected in the rudiment and growth plate chondrocytes, and was found in the perichondrial capillary networks and cartilage canals (Melrose et al, 2004).…”
