Expression of phosphorylcholine-specific B cells during murine development

Abstract: The TEPC 15 (T15) clonotype, a putatively germline antibody specificity, does not appear in the neonatal B-cell repertoire until approximately 1 wk of age. This report extends this observation by the demonstration that (a) the T15 clonotype follows similar kinetics of appearance in germfree as well as conventionally-reared mice; (b) maternal influences and genetic background play a minor role in the development of the T15 clonotype since CBFI neonates raised by C57BL/6 or BALB/c mothers acquire the T15 clonoty… Show more

“…Although the B cell specificity repertoire of adult individuals is extraordinarily diverse, inbred murine strains reproducibly display certain clonotypes at a relatively high frequency (1)(2)(3)(4)(5)(6)(7)(8)(9). Often such clonotypes dominate the in vivo and in vitro response to given antigens.…”

“…Often such clonotypes dominate the in vivo and in vitro response to given antigens. The response to phosphorylcholine (PC) ~ of BALB/c mice is one such response, wherein the repertoire of responsive clonotypes includes over 100 distinct specificities (10,11), yet one clonotype, that which is indistinguishable from the TEPC-15 (T 15) myeloma protein, represents the majority of the response (5)(6)(7)(8)(9)(10)(11). The basis for such clonai dominance within an immune response is not yet fully understood.…”

“…This is in contrast to specificities directed against other haptens such as phosphorylcholine, which develop later in ontogeny (22). From these results Klinman and Press (21) predicted that the DNP clonotypes would be expressed as early as the 15th d of gestation in fetal spleen.…”

“…Considerable evidence indicates that there is a sequential appearance of various specificities or clonotypes (22,26,27), and day 18 or 19 may mark the appearance of many later-developing, non-DNP specificities.…”
