Expression of PCNA and CD44mRNA in colorectal cancer with venous invasion and its relationship to liver metastasis

Abstract: AIM:To investigate the expression of proliferating cell nuclear antigen (PCNA) and CD44mRNA in colorectal cancer with venous invasion and its relationship with liver metastasis. METHODS:Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of PCNA and CD44mRNA in 31 cases of colorectal cancer with venous invasion. RESULTS:Positive expression rates of PCNA and CD44mRNA in colorectal cancer were higher than those without liver metastasis (P<0.05 and P<0.01). In case of colore… Show more

“…Mechanistically, our results show that HMGB1 induces CAC tumorigenesis and PCNA expression through the ERK1/2 signaling pathway. PCNA is a chief proliferation marker that reflects the level of cell proliferation [ 32 , 33 ]. Our finding is consistent with a previous report showing that blocking the RAGE-HMGB1 axis suppresses the growth and metastases of C6 glioma cells by inhibiting activation of the MAPK pathway [ 29 ].…”

“…PCRs involved an initial denaturation at 94°C for 2 minutes and 30 seconds, followed by 25 or 40 cycles at 94°C for 30 seconds, and an appropriate annealing temperature (58°C to 65°C) for 30 seconds and then 72°C for 55 seconds. PCR primer sequences of the examined genes are shown in Table 2 33‐37 . The exponential phases of RT‐PCR amplification runs were determined from 25 to 40 cycles to allow quantitative comparisons among the cDNAs.…”

“…The DNA polymerase cofactor, proliferating cell nuclear antigen (20p12), regulates both synthesis of DNA and cell cycle, and expression of proliferating cell nuclear antigen has been found to associate with liver metastasis of CRC. 51 Adenosylhomocysteinase ( at 20q11) is required for Myc-induced mRNA cap methylation, protein synthesis and cell proliferation. 52,53 In 20q13.12, the transcription factor HNF4A manipulating polarity of epithelial cell showed facilitation in mouse intestine neoplasia.…”
