Expression of p27 is associated with Bax expression and spontaneous apoptosis in oral and oropharyngeal carcinoma

Abstract: p27 Kip1 , a cyclin-dependent kinase inhibitor, is a negative regulator of the cell cycle, and apoptosis is a genetically encoded program of cell death. To clarify the relationship between the cell cycle and apoptosis, we investigated expression of p27, cyclin D1 and apoptosis-related proteins (p53, Bax, Bcl-2 and c-Myc) in 60 cases of oral and oropharyngeal squamous-cell carcinoma (SCC) using an immuno-histochemical approach, and evaluated spontaneous apoptosis in vivo. Our most notable finding was that spon… Show more

“…Several studies have analyzed c-myc expression in this type of tumor showing different results, with an average positivity of 41.28% (2.4-75%) between the different studies (6,(26)(27)(28)(29)(30)(31)(32)(33)(34). The results are extremely contradictory, as well as the variation in the quantification methods.…”

“…This gene regulates G 0 -to-S phase transitions by interacting with cyclindependent kinases (CDKs) (Hengst and Reed 1998;Sherr and Roberts 1999), and overexpression of this gene has been shown to trigger apoptosis in the MDA-MB-231 cell line (Katayose et al 1997) with pro-apoptotic properties in other cancers (Fujieda et al 1999;Wu et al 1999;Tenjo et al 2000). Apoptosis has recently been identified as one of the deregulated pathways by the miR-182-5p cluster in medulloblastoma (Weeraratne et al 2012), and suppression of CDKN1B by miR-182-5p, along with other pro-apoptotic genes like BAK and BAX (Supplemental Table 1), suggests that apoptosis triggered by DNA damage could be deregulated by this miRNA.…”

“…Immunohistochemical staining was performed to detect Syk in human nasal fibroblasts using the traditional avidin-biotin-peroxidase complex technique (29). After purified fibroblasts were seeded and cultured on chamber slide-glasses, the cells were fixed with 70% ethanol.…”
