Expression of p21Waf1/Cip1 and Cyclin D1 Is Increased in Butyrate-resistant HeLa Cells

Abstract: Sodium butyrate induced cell cycle arrest in mammalian cells through an increase in p21 Waf1/Cip1 , although another study showed that this arrest is related to pRB signaling. We isolated variants of HeLa cells adapted to growth in 5 mM butyrate. One of these variants, clone 5.1, constitutively expressed elevated levels of p21 Waf1/Cip1 when incubated in regular growth medium and in the presence of butyrate. Despite this elevated level of p21 Waf1/Cip1 , the cells continue to proliferate, albeit at a slower ra… Show more

“…Upregulation of cyclin D1 and downregulation of p21 and Bax are also involved in BR human CRC cells, HCT116, HT29 and SW480 [ 35 ]. HeLa human cervical cancer cells have also been reported to develop resistance to butyrate via upregulation of cyclin D1 [ 43 ]. Tamoxifen-resistant breast cancer cells, MCF7 and T47D, have been reported to overexpress cyclin D1 owing to the inhibition of cyclin D1 degradation [ 44 ].…”

“…However, when colon cancer cells are continuously exposed to butyrate, anticancer activity is lost as the cells become resistant to butyrate [ 17 , 18 ] and eventually become resistant to butyrate [ 18 ]. BCS-TC2 human colon adenocarcinoma cells [ 19 ] and HeLa human cervical cancer cells [ 20 ] also showed resistance to butyrate.…”
