Expression of nitric oxide synthase isoforms and arginase in normal human skin and chronic venous leg ulcers

Abstract: Chronic venous ulcers, an example of abnormal wound healing, show chronic inflammation with defective matrix deposition which together with the underlying vascular pathology, result in delayed healing. L-arginine is known to be metabolized by one of two pathways: nitric oxide synthase (NOS), producing nitric oxide (NO), or arginase, producing ornithine. NO is involved in many pathological conditions including vascular and inflammatory disorders. This study therefore investigated the distribution, level and act… Show more

“…Immunostaining for iNOS and ecNOS was absent from the negative control in which the primary antibodies were omitted (Figures 3C and 4F). This indicates the specificity of the antibodies used for iNOS and ecNOS, which was also confirmed in previous studies by preā€adsorption with immune serum 12, 13. Results were consistent in both the normal controls and the cirrhotic livers for both antibodies.…”

“…Haematoxylin was used as a counterstain to delineate the nuclei. Previous studies from our group have confirmed the specificity of the iNOS and ecNOS antibodies by the use of positive controls and the finding of negativity using preā€adsorption with immune serum 12, 13.…”

“…Both aspects are widely considered a biologic advantage because while binding iron, it avoids ROS production via the Fenton and Haber-Weiss reactions. 23,24 The main finding of our study is the demonstration of the increased release of iron in the serum of patients affected by severe CVD. This latter condition is readily visible, for example, in patients affected by liver hemochromatosis, but it causes nothing more than a mild dermal atrophy.…”

“…It is our contention the effects observed in this case series is not surprising in light of the mechanism by which HWDS increases microcirculation in rat experiments [7]. Moreover, nitric oxide is profoundly involved with wound healing by virtue of its effects to control blood flow to tissues and anti-inflammatory effects to influence pain [11]. Interestingly, wound healing represents a particularly challenging clinical problem to which no efficacious treatment regimens currently exist.…”
