IntroductionObesity exacts a continuing toll on public health and is a wellknown risk factor for diabetes, hypertension, hyperlipidemia, and cardiovascular diseases [1][2][3][4]. It is evident that not only hypertrophy of adipocytes and increase of fat deposit but also proliferation of preadipocytes and differentiation from pre-adipocytes to adipocytes have important roles for forming obesity. The most intensive and clinical interventions to prevent obesity have been primarily directed to decrease excessive amounts of fat tissue by a change in the balance between intake and expenditure of energy using daily exercise and diet control. On the other hand, the inverse correlation between body mass index (BMI) and the average temperature of January exists, and BMI is low when the temperature is high [5]. In addition, when rats were fed under low or high temperature environment, the amount of subcutaneous fat in rats fed under high temperature was significantly low, compared with that under the low temperature [5]. Thus, hyperthermia may affect adipose cell functions, then adipogenesis, and provides a therapeutic or preventive modality against obesity-related diseases. However, there are few reports concerning the direct effects of hyperthermia stimulation on adipocytes and its underlying mechanism. AbstractBackground: Hyperthermia may affect adipose cell functions and then adipogenesis. Thermo-sensitive transient receptor potential (TRPs) proteins such as TRPV1-4 play vital roles in cellular functions. We investigated the expression of thermo-sensitive TRPVs and their roles on IL-6 production in mouse 3T3-L1 preadipocytes and differentiated adipocytes.

Brown and beige adipocytes are a major site of mammalian non-shivering thermogenesis and energy dissipation. Obesity is caused by an imbalance between energy intake and expenditure and has become a worldwide health problem. Therefore modulation of thermogenesis in brown and beige adipocytes could be an important application for body weight control and obesity prevention. Over the last few decades, the involvement of thermo-sensitive transient receptor potential (TRP) channels (including TRPV1, TRPV2, TRPV3, TRPV4, TRPM4, TRPM8, TRPC5, and TRPA1) in energy metabolism and adipogenesis in adipocytes has been extensively explored. In this review, we summarize the expression, function, and pathological/ physiological contributions of these TRP channels and discuss their potential as future therapeutic targets for preventing and combating human obesity and obesity-related metabolic disorders.

Effects of treadmill running and rutin on lipolytic signaling pathways and TRPV4 protein expression in the adipose tissue of diet-induced obese mice

To explore the effects of rutin and exercise on high-fat diet (HFD)-induced disrupted lipolytic signaling, adenosine 5'-monophosphate (AMP)-activated protein kinase (AMPK) signaling, transient receptor potential cation channel subfamily V member 4 (TRPV4) and its associated protein expression, and whether depot-specific effects existed. C57BL/6J mice were randomized into five groups: chow group, HFD, HFD plus rutin intervention group (HR), HFD combined with treadmill running group (HE), and HFD combined with treadmill running and rutin intervention group (HRE). At the end of the 16-week intervention, lipolytic markers, AMPK signaling pathways, TRPV4, and peroxisome proliferator-activated receptor gamma coactivator 1α + β (PGC-1α + β) from adipose tissue were measured by western blotting. In epididymal adipose tissue, HFD resulted in significant reduction in the phosphorylation of hormone sensitive lipase at serine660 (p-HSL660), perilipin A, phosphoenolpyruvate carboxykinase (PEPCK), p-AMPK, and p-acetyl-CoA carboxylase (ACC) protein expression. Exercise intervention and exercise plus rutin completely restored p-HSL660, perilipin A, PEPCK, p-AMPK, and p-ACC protein expression to normal level. HFD and HR groups have reduced expression of PGC-1α + β, exercise, and exercise plus rutin completely restored PGC-1α + β expression to normal level. In subcutaneous adipose tissue, HFD elevated TRPV4, exercise, and exercise plus rutin completely reduced TRPV4 to normal level. HR, HE, and HRE group have increased PGC-1α + β. In conclusion, depot-specific effects existed in regards to how rutin and exercise affect lipolytic signaling and p-AMPK, as well as TRPV4 and PGC-1α + β expression.
