Expression of microRNAs in the ascites of patients with peritoneal carcinomatosis and peritonitis

Abstract: The data from the current study provide novel evidence of the differential expression of miRNAs in ascites from patients with PCA and SBP, which may offer an additional miRNA-based molecular approach for the differential diagnosis of PCA. “…Subsequent steps were as described above. The miRNA extraction from ascites is similar to serum and is described in detail previously . In total nine samples were used to evaluate the detectability of xenomiR miR‐168 in ascites.…”

“…Deregulation of miRNAs is an early epigenetic event and therefore potentially reversible . Certain miRNAs, for instance miR‐21, are frequently deregulated in GI tumors and are suggested as potential tumor biomarkers . We and others have shown that miRNAs can be easily detected in feces and differential expression of miRNAs may be useful for diagnosis of CRC and inflammatory bowel diseases …”

“…Once the exosomes have been isolated, many commercially available kits can be used for RNA extraction. A recent study by Schindler et al of 3 different extraction methods has indicated that the extraction method can have an impact on the amount of RNA extracted from ascitic fluid specimens. After RNA extraction, miRNA quantification may be achieved by real‐time polymerase chain reaction systems or by next‐generation sequencing analysis.…”

“…Implants often are from ovarian malignancies and less frequently are from the gastrointestinal tract. Schindler et al compared miRNA expression profiles in patients with peritoneal carcinomatosis, spontaneous bacterial peritonitis (SBP), and portal hypertension without peritonitis and demonstrated a significant upregulation of miRNA‐21 and miRNA‐186 in patients with carcinomatosis versus the other groups. Overexpression of miRNA‐223 was found in SBP; this finding is not surprising because this miRNA is highly expressed in acute inflammatory cells.…”

“…Schindler et al prospectively analysed samples from patients with peritoneal carcinomatosis (PCA), spontaneous bacterial peritonitis (SBP), and portal hypertension (no SBP/PCA). As a result, a simple proportion score between miR-21 and miR-223 allowed to discriminate between the patients with PCA and those with SBP with an area under the curve of 0.982 (95% confidence interval, 0.943-1.022) [4]. Circulating miRNAs are also proposed as a biomarker of heart disease.…”
