Expression of kindlin-3 in melanoma cells impedes cell migration and metastasis

Abstract: Kindlins are a small family of 4.1-ezrin-radixin-moesin (FERM)-containing cytoplasmic proteins. Kindlin-3 is expressed in platelets, hematopoietic cells, and endothelial cells. Kindlin-3 promotes integrin activation, clustering and outside-in signaling. Aberrant expression of kindlin-3 was reported in melanoma and breast cancer. Intriguingly, kindlin-3 has been reported to either positively or negatively regulate cancer cell metastasis. In this study, we sought to clarify the expression of kindlin-3 in melanom… Show more

“…For Kindlin-2, increased levels were reported to enhance tumor invasion and poor prognosis in breast, bladder, pancreas, stomach cancers and malignant mesothelioma, whereas they correlated with reduced tumor invasion and metastasis in colorectal and serous epithelial ovarian cancers [ 14 , 18 23 ]. Moreover, recent studies provided conflicting results suggesting either a tumor suppressor or a tumor promoting activity of Kindlin-3 in breast cancer and melanomas [ 14 , 24 26 ].…”

“…However, independent evidence for the role of kindlin-3 in BC can be taken from interrogation of the Oncomine database (http://www.oncomine.com) in which kindlin-3 ranked in the top 3% of up-regulated genes. Furthermore, in glioblastoma, kindlin-3 contributed to resistance to telozolomide and cell proliferation through Wnt signaling (Lu et al, 2017), whereas overexpression of kindlin-3 in melanoma cells led to imbalanced Rho GTP-ase activation and inhibition of cell migration (Feng et al, 2017).…”
