Expression of Human Calcitonin by Microencapsulated Recombinant Myoblasts

Abstract: A novel approach to treat post-menopausal osteoporosis is proposed by engineering cell lines for the secretion of human calcitonin (hCT) and which would then be suitable for implantation in different allogeneic hosts. Mouse myoblast were transfected with the cDNA for hCT using the liposome-based gene delivery technique and clones secreting of human calcitonin were isolated. Human calcitonin expression was detected by ELISA. Western blot and immunohistochemical analyses also confirmed the plasmid, pcDNA3-hCT, h… Show more

“…43 In another study, investigators compared the toxic effects of actinomycin-D encapsulated either in the aqueous phase or in the lipid phase of liposomes and the nonencapsulated actinomycin-D on the blood forming system on cell proliferation in the intestine and on antibody production by spleen lymphocytes and have confirmed that actinomycin-D is less toxic to mice than nonencapsulated actinomycin-D, but retains its tumoricidal activity. 44 Similarly, nerve growth factor has also been encapsulated into liposomes in order to protect it from the enzyme degradation in vivo and promote permeability across the blood-brain barrier. 45 Furthermore, MEND has been developed as a novel nonviral gene delivery system.…”

“…Gene therapy has emerged as one of the most rapidly advancing fields of biotechnology with great promise for treating various diseases (Jung et al 2005;Wang et al 2006). Bone regeneration needs to be enhanced when treating fracture non-unions and healing large bone defects due to tumor or trauma.…”

“…Synthetic CT is a therapeutic peptide widely used for treating osteoporosis and CT produces analgesia with a mechanism different from opiate and is effective in patients with refractory pain (Reale et al 2001;Wang et al 2006). However, the major disadvantages of CT therapy are its high cost and low yield.…”

“…Two termination codons at the end of the 5hsCT gene were enough to end the translation of the 5hsCT gene (Wang 2005). The Ura3 gene used as an auxotrophic selection marker was equipped with its own promoter and was independently located in front of the expression cassette.…”
