Expression of high levels of nitrobenzylthioinosine-sensitive nucleoside transport in cultured human choriocarcinoma (BeWo) cells

Abstract: We have examined binding of [3H]nitrobenzylthioinosine (NBMPR) and influx of [3H]thymidine in adherent cultures of human choriocarcinoma (BeWo) cells and, for comparison, cervical-carcinoma (HeLa) cells. Specific association of NBMPR with BeWo cells at 22 degrees C required 1.5 h to reach an equilibrium between free and bound ligand, whereas association with HeLa cells required 20-30 min. Scatchard analysis of NBMPR binding to low-density cultures of BeWo cells revealed a total of 27 x 10(6) sites per cell, co… Show more

“…2B, Table II) exhibited K d and B max values of 0.9 Ϯ 0.2 nM and 75.0 Ϯ 5 pmol/mg of protein, respectively (mean Ϯ S.E., n ϭ 4). The K d value was essentially the same as that determined previously (0.6 nM) for the high affinity binding sites in intact BeWo cells (19), and there was no indication of a second set of lower affinity binding sites. There was a nearly 4-fold increase in specific binding activity relative to that of the crude membrane preparations ( Fig.…”

“…BeWo Cells Express hENT1 and hENT2 mRNA-BeWo cells have previously been shown (19) to exhibit es and ei functional transport activity. Since hENT1 and hENT2 were initially identified by molecular cloning of a placental cDNA library (10,16) and the BeWo cell line is of placental origin (18), BeWo RNA was examined for evidence of expression of hENT1 and hENT2 mRNA.…”
