Expression of H-Y Antigen in Human Males with Two Y Chromosomes

Abstract: To determine whether the gene that controls the expression of H-Y ("male") antigen on human cells is Y-linked, we have compared the H-Y antigen level in normal males with that in three males with two Y chromosomes. Leukocytes from one XXYY and two XYY males express more H-Y antigen than leukocytes from normal XY males. We conclude that a structural gene or positive regulatory gene for H-Y antigen is on the human Y chromosome. Testing for the H-Y antigen may be of benefit in patients who have signs of masculini… Show more

“…In the standard cytotoxicity test, mouse sperm were incubated for 50 minutes with both H-Y antiserum and rabbit complement. Trypan-blue dye was added during the last 10 minutes of incubation to stain dead sperm (44). the mouse, she screened all available mouse antiserums to find out what known antigens might be expressed on sperm, including, in her survey, serums from B6 female mice that had rejected grafts from B6 males.…”

“…Until recently, there existed no genetic markers for the Y chromosome and thus no way to correlate chromosome structure with male function. This situation is now changing with the discovery of two distinctive cytological characteristics on the human Y chromosome (3,4) and the recognition that a locus for a male histocompatibility antigen is Y-linked (5,6).…”

“…Duplication of the short arm, as in the subject of the present study, is associated with increased expression of H-Y relative to that in normal males.
