Expression of Cytochrome P450 Genes in CD34 + Hematopoietic Stem and Progenitor Cells

Abstract: Expression of major cytochrome P450 forms (P450) was followed in preparation of purified hematopoietic CD34 + stem and progenitor cells. Levels of transcripts as well as mature proteins were traced by quantitative real-time polymerase chain reaction and by Northern and Western blotting. P450 1B1 and P450 2E1 proteins and respective mRNAs were found in all cases. On the other hand, no expression of P450 3A4, P450 3A7, and P450 2C9 was found. The results showed that expression of various P450 enzymes starts at d… Show more

“…Recent study demonstrated the expression of several CYP genes in the CD34 ϩ hematopoietic stem cells. 23) Together with our data, it is possible that the cells at the early stage of hematopoietic differentiation may be influenced by interaction with xenobiotics, depending on the extents of CYP expresstion.…”

“…Various results have shown that hematopoietic stem and progenitor cells express high levels of ABC transporters [72,73] as well as CYP1B1 [74]. Indeed, strong expression of CYP1B1 mRNA and protein have been found in purified hematopoietic CD34 þ undifferentiated cells from two patients' peripheral blood [74]. These latter results underline that imatinib metabolism should also be considered in these undifferentiated cells.…”

“…Moreover, imatinib efficacy appears to be reduced in stem and progenitor cancer cells [69][70][71] and their survival (quiescent cells) seems to be the starting point for the emergence of resistant cell clones and treatment relapse [71]. “…1A, f). Although cyanide and SA are not highly specific inhibitors of MPO but rather block hemecontaining enzymes and total heme synthesis, respectively, together, these results strongly suggest that H 2 O 2 -dependent MPO catalysis is involved in the generation of etoposide-O ⅐ radicals, especially because MPO is a major heme-containing protein in growth factor-mobilized CD34 ϩ cells and there is little expression of cytochromes P450 in CD34 ϩ cells capable of etoposide oxidation (Soucek et al, 2005).…”
