Expression of CC Chemokine Receptor-7 and Regional Lymph Node Metastasis of B16 Murine Melanoma

Abstract: Expression of a single chemokine receptor gene, CCR7, increased B16 cell metastasis to draining lymph nodes, suggesting that cancer cells may co-opt normal mechanisms of lymph node homing during metastasis.

“…For instance CXCL12, the ligand for CXCR4, is strongly expressed in lung, liver, bone marrow and lymph nodes while CCL21, the ligand for CCR7, is strongly expressed in lymph nodes [52]. In a melanoma model, transfection of CCR7 promotes lymph node metastases while transfection of CXCR4 promotes lung metastases [53,54]. These data indirectly support the absence of relationship between axillary lymph node involvement and detection of circulating breast cancer cells.…”

“…It was also reported that Caco 2 metastasized to LNs at a low frequency (1/10) with a cecal CXCR3 in colon cancer metastasis to lymph nodes K Kawada et al transplantation model (Flatmark et al, 2004). Recent studies showed that CCR7 was involved in LN metastasis in breast and stomach cancers, and melanoma (Mu¨ller et al, 2001;Wiley et al, 2001;Mashino et al, 2002). Among the colon cancer cell lines tested by RT-PCR and flow cytometry, however, we found expression of CCR7 only in SW480 and Caco2 that rarely metastasized to LNs (Figure 1b and c).…”

“…breast, melanoma, gastric, non-small cell lung cancer, head and neck tumors, colorectal carcinoma), CCR7 is upregulated and mediates tumor cell dissemination to lymph nodes [101][102][103][104][105][106]. In a recent study, brain infiltration by T-cell leukemic blasts was mediated by CCR7 [15].…”
