Expression of botulinum neurotoxins A and E, and associated non-toxin genes, during the transition phase and stability at high temperature: analysis by quantitative reverse transcription-PCR

Abstract: Production of botulinum neurotoxin A (BoNT/A) and associated non-toxic proteins (ANTPs), which include a non-toxic non-haemagglutinin (NTNH/A) as well as haemagglutinins (HAs), was found previously to be dependent upon an RNA polymerase alternative sigma factor (BotR/A). Expression of the botR/A, bont/A and antp genes, monitored by reverse transcription and real-time PCR analysis, occurred concomitantly at the transition between the exponential and stationary growth phases of Clostridium botulinum A. The botR/… Show more

“…Although data for cntC are not reported, relative expression is anticipated to be similar to that of the other hemagglutinin-encoding genes (cntD and cntE), since all three form an operon regulated by the alternative sigma factor CntR. Indeed, their cotranscription has been reported by Couesnon et al (10). Expression of the regulatory gene, cntR, was consistent with the remainder of the neurotoxin gene cluster but at a lower rate, as observed previously (10).…”

“…TEM micrographs of several type E strains did not consistently show morphologically distinct filaments, suggesting that FlaA1/A2 and FlaB may be found in the same filament. Type E strains possess other unique characteristics, including the mechanism of regulation of the type E BoNT (9).…”

“…Expression and Purification of Recombinant Proteins-The BoNT isoform A holotoxin was purified as previously described (21). The E. coli strain XL1Blue (Novagen, Madison, WI) was used as the host for recombinant proteins productions.…”
