“…The PVDF membrane was blocked in 5% (wt/vol) nonfat milk, pH 7.4, in 1ϫ PBS containing 0.05% Tween-20 (PBST) for 30 min and probed with appropriate primary antibodies. Rabbit polyclonal anti-placental alkaline phosphatase antibody, which recognizes the unmodified and modified placental alkaline phosphatases, was used at 1/2,000 overnight at 4°C (76), and mouse anti-cadherin-11 antibody at 1/1,000 overnight at 4°C in blocking buffer. The membrane was washed with PBST, probed with HRP-conjugated, appropriate secondary antibody at 1/10,000 for 1 h at room temperature, and detected using Enhanced Chemi-Luminiscence reagent according to manufacturer's instructions.…”
