Expression of ACTH-induced corticosteroid biosynthesis in newborn rat adrenocortical cells cultured in serum-free and carrier protein-free medium containing a cholesterol-?-cyclodextrin complex

Abstract: Newborn rat adrenocortical cells were successfully cultured in a serum-free carrier protein-free medium (SPFM) by using alpha-cyclodextrin as a cholesterol carrier and have expressed corticosteroid biosynthesis in this medium. A stable inclusion complex of cholesterol-alpha-cyclodextrin with a molar ratio of almost 1 was obtained for a 5 X 10(-5) mol/1 alpha-cyclodextrin concentration. Cell cultures incubated with [4-14C] cholesterol-alpha-cyclodextrin in SPFM produced, under ACTH stimulation, various 14C labe… Show more

“…f 0 Spectra were recorded as described in note a, Table I, and are of a multilayer sample prepared on the initial monolayer presented in Table I as sample 1. 'The notation [1][2][3][4][5][6][7] means the spectrum of the seven-layer sample; layer eight is the spectrum of the eighth layer obtained by subtraction of the seven-layer spectra from the eight-layer spectra. ' The samples were either those with an ester surface (E) or reduced (R) to the alcohol surface.…”

“…Delivery and supply of lipids have been shown to be an important role of serum and one of the most difficult functions to replace in chemically defined media. Cyclodextrins, which are cyclic glucose compounds with a hydrophobic interior and a hydrophilic exterior, have been utilized as carriers for fatty acids and cholesterol with some success in serum-free medium formulations (Gorfien et al, 2000;Greenberg-Ofrath et al, 1993;Hammami et al, 1986;Keen and Steward, 1995;Minamoto et al, 1991). Pluronic 1 F-68 (PF-68), which is commonly used in cell culture medium to protect cells from bubble damage (Handa-Corrigan et al, 1989) has also been used to emulsify lipids to allow delivery into aqueous culture media (Maiorella et al, 1988).…”
