Expression Cloning and Pharmacological Characterization of a Human Hippocampal Neuropeptide Y/Peptide YY Y2 Receptor Subtype

Abstract: The pancreatic polypeptide family includes neuropeptide Y (NPY), one of the most abundant neuropeptides in the mammalian nervous system, as well as peptide YY (PYY) and pancreatic polypeptide (PP). This peptide family is involved in numerous physiological processes such as memory, pain, blood pressure, appetite, anxiety, and circadian rhythm. Of the multiple Y-type receptors proposed for PP family members, only the Y1 subtype was cloned previously. We now report the isolation of a human Y2 (hhY2) receptor cDNA… Show more

“…The library was plated on agar plates (ampicillin selection) in 584 primary pools of ϳ4,500 independent clones. Preparation and transfection of library pool DNA were carried out as described (23). Forty-eight h after transfection, the galanin binding assay was carried out with 1 nM 125 I-porcine galanin ( 125 IpGAL; specific activity ϳ2200 Ci/mmol; NEN Life Science Products) in 20 mM HEPES-NaOH, pH 7.4, containing 1.26 mM CaCl 2 , 0.81 mM MgSO 4 , 0.44 mM KH 2 PO 4 , 5.4 mM KCl, 10 mM NaCl, 0.1% bovine serum albumin, and 0.1% bacitracin for 1 h at room temperature.…”

“…A low-phase Fouriertransform filter was used to reduce high-frequency noise. The ellipticity was expressed as mean-residue molar ellipticity [Q] R in deg´cm 2´d mol 21 . The secondary structure components were calculated by the secondary structure estimation Program (JASCO), based on the reference spectra of Yang [42].…”

“…PP binds primarily to the Y4 receptor 7 and PYY 3À36 binds predominantly to the Y2 receptor. 8 In addition to their central actions, PP and PYY may also inhibit food intake through a reduction in gut motility. 9,10 There is evidence that co-administration of appetiterelated peptides may inhibit feeding synergistically.…”
