SummaryThe complement system is an important part of the innate immune defence. It contributes not only to local inflammation, removal and killing of pathogens, but it also assists in shaping of the adaptive immune response. Besides a role in inflammation, complement is also involved in physiological processes such as waste disposal and developmental programmes. The complement system comprises several soluble and membrane-bound proteins. The bulk of the soluble proteins is produced mainly by the liver. While several complement proteins are produced by a wide variety of cell types, other complement proteins are produced by only a few related cell types. As these data suggest that local production by specific cell types may have specific functions, more detailed studies have been employed recently analysing the local and even intracellular role of these complement proteins.Here we review the current knowledge about extrahepatic production and/ or secretion of complement components. More specifically, we address what is known about complement synthesis by cells of the human immune system.

Background: Previous studies introduced ORZ for pharmacological purposes. As ORZ reduces NKs originated IFN-γ potentially. Therefore this study was designed to evaluate VEGF and MMP-9 expression in HEP-G2 cell line after culture with the soup of ORZ treated PBMCs. Materials and methods:In current in-vitro study, PBMCs from seven healthy male subjects (aged 20 to 30 years) were isolated by ficoll density gradient. The soup of ORZ treated PBMCs were cultured with HEP-G2 cell line and the VEGF and MMP-9 expression in the cancerous cell line were measured by real time PCR.Results: VEGF and MMP-9 expression increased after 48 hours, but only the increment of MMP-9 expression was statistically significant (P<0.05). The expression of these genes did not alter significantly after 24h and 72h incubation of the cancer cells with the soup of ORZ treated PBMCs soup (P>0.05). Conclusion:By considering the increase of MMP-9 and VEGF expression in the cancerous cell line cultured with ORZ-stimulated PBMCs soup, it seems long term administration of ORZ, as a drug, may change the response of immune system toward carcinogenesis.

Complement-dependent cytotoxicity (CDC) has been suggested to be an important mechanism of action of tumor-targeting Abs. However, single unmodified epidermal growth factor receptor (EGFR)–targeting IgG1 Abs fail to trigger efficient CDC. For the current study, we generated a CDC-optimized variant of the EGFR Ab matuzumab (H425 wt) by introducing amino acid substitutions K326A/E333A (H425 mt). This Ab was then used to elucidate the impact of complement activation on the capacity of effector cells such as mononuclear cells (MNC) and polymorphonuclear cells (PMN) to exert Ab-dependent cell-mediated cytotoxicity (ADCC). H425 mt, but not H425 wt, significantly induced complement deposition, release of anaphylatoxins, and CDC against distinct tumor cell lines, whereas no differences in ADCC by MNC or PMN were detected. Notably, stronger cytotoxicity was induced by H425 mt than by H425 wt in whole blood assays and in experiments in which MNC or PMN were combined with serum. Although MNC-ADCC was not affected by C5 cleavage, the cytotoxic activity of PMN in the presence of serum strongly depended on C5 cleavage, pointing to a direct interaction between complement and PMN. Strong cell surface expression of C5a receptors was detected on PMN, whereas NK cells completely lacked expression. Stimulation of PMN with C5a led to upregulation of activated complement receptor 3, resulting in enhanced complement receptor 3–dependent PMN-ADCC against tumor cells. In conclusion, complement-optimized EGFR Abs may constitute a promising strategy to improve tumor cell killing by enhancing the interaction between humoral and cellular effector functions in Ab-based tumor therapy.
