Expression and localization of Nlrp4g in mouse preimplantation embryo

Abstract: The Nlrp gene family contains 20 members and plays a pivotal role in the innate immune and reproductive systems in the mouse. During evolution, seven Nlrp4 gene copies (named from Nlrp4a to Nlrp4g). Nlrp4a-Nlrp4g have arisen that display specific or preferential ovarian expression patterns. However, the expression pattern and localization of Nlrp4g in mouse preimplantation embryo development are unknown. Here we report that Nlrp4g was highly expressed in mature oocytes and zygotes, then downregulated and not d… Show more

“…After ovulation and fertilization, the NLRP9B protein was present throughout early embryogenesis, while the protein level diminished gradually with embryo development. This expression pattern was similar to those of NLRP2, NLRP4G and NLRP5 during preimplantation embryo development [ 6 , 8 , 16 ]. Confocal microscopy revealed that the NLRP9B protein was predominantly localized in the cytoplasm of blastomeres.…”

“…Confocal microscopy revealed that the NLRP9B protein was predominantly localized in the cytoplasm of blastomeres. This distribution of NLRP9B protein is similar to those of NLRP2 and NLRP4G [ 6 , 16 ] and differs from that of NLRP5, which is mainly located in the subcortical regions of oocytes and preimplantation embryos [ 3 , 4 ].…”

“…Mice and humans have 14 and 20 NLRP-encoding genes [5][6][7], respectively, with a subset being expressed in oocytes and early preimplantation embryos. Of the 14 human NLRP genes, ten NLRPs (2,4,5,7,8,9,11,12,13,14) were detected in oocytes and/or early embryos [7]. Mutations in NLRP genes are associated with different disorders, including developmental diseases [3].…”

“…Once activated, NLRP1 specifically recruits and activates caspase‐1 downstream, which subsequently cleaves pro‐IL‐1β and pro‐IL‐18 and promotes the maturation and release of IL‐1β and IL‐18 (Martinon et al, ). Several lines of evidence show that NLRP2, NLRP4, NLRP5, and NLRP7 play important roles in embryonic development (Bauernfeind et al, ; Murdoch et al, ; Peng et al, ; Zhang et al, ). NLRP3 is the most‐studied member of the NLR family of proteins.…”
