Expression and localization of collagen type IV α1 chain in medaka ovary

Abstract: A cDNA clone coding for collagen type IV α1 chain was obtained from the ovary of the medaka, Oryzias latipes. The clone encodes a protein of 1639 amino acids including a putative 21-residue signal peptide, and the deduced amino acid sequence of the α1 chain was homologous to those of the proteins from other species. Collagen type IV α1 chain mRNA was expressed in various tissues of the adult fish. In situ hybridization analysis revealed that the α1 chain mRNA was localized in the follicle layer of all growing … Show more

“…Moreover, we observed that the collagen IVα5 gene is also expressed at the periphery of the ovarian lamellae during early ovarian differentiation (unpubl. data), and collagen IV is known to be expressed by theca cells in the medaka ovary [Kato et al, 2010]. These similarities between rainbow trout and medaka suggest that the cells expressing both cyp19a1a and fst during rainbow trout early ovarian development are also precursors of theca cells.…”

“…2E). We also analyzed the transcripts of gelatinase B ( mmp9 ), a marker for granulosa cells of the large follicles [25], and collagen type I α1-chain and collagen type IV α1-chain, markers for theca cells [26], [27]. A fragment of mmp9 , but not of collagen type I α1-chain or collagen type IV α1-chain, was amplified, confirming that lhcgrbb mRNA, but not fshra mRNA, is expressed in the granulosa cells of the large follicles.…”

“…It is generally accepted that ECM proteins present in the follicle layers of ovulating follicles are collagen type I, which is associated with the theca cell layer, whereas collagen type IV and laminin are two major components of the follicle basement membrane. Our recent studies have indicated that collagen type I [32] and collagen type IV [33] are localized in the follicle layer of the follicle in the medaka. In addition, gelatinase A was activated by Mmp14 at the plasma membrane of the oocyte to hydrolyze collagen type IV, whereas Mmp15 degraded collagen type I [17].…”
