Expression and Intracellular Localization of Protein Phosphatases 2A and 2B, Protein Kinase A, A-Kinase Anchoring Protein (AKAP79), and Binding of the Regulatory (RII) Subunit of Protein Kinase A to AKAP79 in Human Myometrium

Abstract: Human postmenopausal and pregnant (nonlabor and labor) myometrium expressed PP2A, PP2B, PKA, AKAP79, and PKA (RII)-AKAP79 binding. Levels of PP2A, PKA, and AKAP79 expression did not appear to be determinants of human myometrial contractility at parturition. Expression of PP2B may play a role in uterine quiescence. No association was found between protein expression and hormone level.

“…The molecular weight estimation by SDS PAGE of approximately 44 kDa for PKAα Cat is the same reported by the antibody manufacturer when detecting PKAα Cat in mouse brain extract ( ) and similar to the estimated molecular weight of PKAα Cat previously found in the cytoplasm of human myometrium (approximately 42 kDa) [ 8 ]. SDS PAGE has an accuracy in molecular weight determination of up to approximately +/- 10% [ 26 ].…”

“…There are several forms of AKAP (often differentiated by a suffix corresponding to their molecular weight) that have been found in tissues and species other than placenta and human. The only form of AKAP found in human reproductive tissue prior to this investigation was AKAP79 which, was shown to be present in the cytoplasm of human myometrium and is thought to anchor PKA to the myometrial plasma membrane [ 8 ].…”

“…Progesterone treatment to postpone labor prevented the decline of membrane PKA 5. Data in humans also support a decline in plasma membrane‐associated PKA, as well as transcriptional downregulation of PKA, between not‐in‐labor and in‐labor samples 6,7. These data indicate that PKA acts as a functional counteragent to myometrium contraction.…”

“…) and may play a similar role in the myometrium (Ayres et al . ). Another possible explanation for the decline in HSP20 Ser16‐phosphorylation may be attributed to 8‐bromo‐cAMP being able to activate PKG when used at high concentrations (as discussed above) and thus negative feedback on HSP20 Ser16‐phosphorylation may have originated from the prolonged activation of PKG instead of altered PKA activity; the same possibility could also apply to the loss of HSP20 Ser16‐phosphorylation following prolonged exposure to 6‐Bnz‐cAMP.…”
