Expression and immunogenicity of recombinant glycoprotein D of herpes simplex virus 1 in Drosophila S2 cells

Abstract: Herpes simplex virus type 1 (HSV-1) is responsible for cold sores in the general population, but also contributes to the development of other more serious diseases in some circumstances. The viral glycoprotein D (gD) is essential for virus entry into host cells. In the present study, the Drosophila melanogaster Schneider 2 (S2) expression system (DES) was evaluated for the expression of recombinant gD1. The DNA sequences encoding the full-length gD1 (369aa, FLgD1) and a truncated gD1 form corresponding to the … Show more

“…While gp120 was obtained based on previous 10.3389/fmicb.2023.1136664 Frontiers in Microbiology 03 frontiersin.org work (Hollister et al, 2014). A Readytoprocess WAVE 25 biological reaction system (GE Healthcare; Boston, USA) was used to grow Drosophila S2 cells for HIV-1 VLP protein production as previously described in our lab with some minor adjustments (Mao et al, 2016). Specifically, 1.2 × 10 7 S2 cells were seeded in a 2-liter bag with 300 mL SFM medium for 3 days culture.…”

“…Results of experiments performed by Mao et al showed that the gD1 ectodomain is secreted while the full-length protein is restricted to the lipid membrane. Mice immunized with the purified gD1 ectodomain generated high levels of antibodies, proving that S2 cells can synthesize immunogenic viral proteins [ 128 ].…”
