Exposure to pressure stimulus enhances succinate dehydrogenase activity in L6 myoblasts

Abstract: .-Contraction of skeletal muscle generates pressure stimuli to intramuscular tissues. However, the effects of pressure stimuli, other than those created by electricity or nerve impulse, on physiological and biochemical responses in skeletal muscles are unknown. The purpose of this study is to examine the effects of a pure pressure stimulus on metabolic responses in a skeletal muscle cell line. Atmospheric pressure was applied to L6 myoblasts using an original apparatus. Succinate dehydrogenase (SDH) activity w… Show more

“…Non-pressurized control HASMCs were settled in a 5% CO 2 incubator at 37°C in the presence of 10mM HEPES. In previous investigations at non-pulsatile atmospheric pressure with biochemical parameters including pH [5], pO 2 and pCO 2 levels maintained at constant levels during the culture [7], cell shape did not detectably change in the HASMCs [8]. The real-time internal pressure of the pressure-loading apparatus was measured (DS-3210 Fukuda denshi, Japan) and recorded by a thermal recorder (RF-85 Fukuda denshi, Japan).…”

“…Depletion of ATP content was observed in STZ-treated cells as a consequence of impaired mitochondrial respiratory chain activity. Succinate dehydrogenase is a mitochondrial TCA cycle enzyme and its activity is mainly regulated by ATP [52]. KIOM-4 treatment restored the ATP content and the succinate dehydrogenase activity, which were both reduced by STZ treatment.…”
