Exploring factors affecting anther culture in rice (Oryza sativa L.)

Abstract: Anther culture is an important in vitro culture technique for the production of double haploids. It is largely species and genotype specific. Asian-cultivated rice (Oryza sativa, ssp. indica) is recalcitrant to anther culture which limits its practical application in rice breeding. Several researchers tried to optimize the medium recipes and culture techniques for callus induction and plantlet regeneration. Negligible response to callus induction and recovery of a higher frequency of albino plants are the majo… Show more

“…These results demonstrated the efficiency of the regeneration medium used. Effect of genotype on plant regeneration from callus cultures have been reported previously (Mon et al, 2020;Sharma et al, 2018;Tripathy et al, 2019). Bagheri andJelodar (2008), andEl-Hennawy et al (2011) reported that plant regeneration from callus was found to be highly heritable.…”

“…Albinism or chlorophyll-deficient regeneration is one of the most important challenges in anther cultures of many cereal crops like barley (Makowska and Oleszczuk 2014), triticale (Krzewska et al 2015), rice (Mishra and Rao 2016), and wheat (Weigt et al 2019). In rice, the frequency of albino plant varies from 5% to 100%, and it is affected by several factors such as the length of cold pre-treatment exposure, genotype, culture medium, and gelling agent (Mishra and Rao 2016;Tripathy et al 2019). The study showed a high frequency of albino plant ranging from 67% in 200 Gy treatment to 100% in 0 Gy and 300 Gy treatments.…”

“…Na geração de plantas haploides, uma das técnicas utilizadas é o cultivo de anteras (TRIPATHY et al, 2019) para a indução de calos e posterior regeneração dos indivíduos haploides. Os calos têm por base uma massa de proliferação celular, que é induzida em um explante inicial por meio da adição de reguladores vegetais, como auxinas, giberelinas e citocininas, que promovem uma desdiferenciação dos tecidos vegetais (MOSTAFA et al, 2020).…”
