Exploring cancer metabolism using stable isotope-resolved metabolomics (SIRM)

Abstract: Metabolic reprogramming is a hallmark of cancer. The changes in metabolism are adaptive to permit proliferation, survival, and eventually metastasis in a harsh environment. Stable isotope-resolved metabolomics (SIRM) is an approach that uses advanced approaches of NMR and mass spectrometry to analyze the fate of individual atoms from stable isotope-enriched precursors to products to deduce metabolic pathways and networks. The approach can be applied to a wide range of biological systems, including human subjec… Show more

“…These derive in part from glycine, which can be made via the glycolytic 3PGA through serine to glycine using SHMT1,2. Additional carbons may derive ultimately from glycine via methylene tetrahydrofolate, which may include a mitochondrial step …”

“…The crucial role of glucose in various metabolic pathways makes 13 C‐labeled glucose derivatives and 13 C MRS, often in conjugation with mass spectrometry, the obvious choice for monitoring glycolysis in vitro as well as in vivo . In an early study, [1‐ 13 C]‐D‐glucose was given to yeast, Candida utilis , to follow glucose metabolism by detecting labeled metabolic products .…”

“…In den letzten Jahren sind durch den Einsatz isotopenmarkierter Tracerverbindungen verschiedene Stoffwechselwege aufgeklärt worden, die in Tumoren unter unterschiedlichen Bedingungen aktiviert werden. Daraus lassen sich wichtige Informationen für die Entwicklung neuer Therapiestrategien gewinnen …”
