Siddha medicine originated from Tamil culture in southern India. Worldwide there are 75% of herbal plants are used for AYUSH and Allopathy medicines. Amukkara Choornam is a polyherbal Siddha formulation. Amukkara Choornam has anti-arthritic activity, and it helps to cure rheumatoid arthritis. The demand for Siddha medicines in India is increasing daily; we have to overcome some problems like quality control issues, processing, administrative issues, and clinical trials. From the above, quality control issues are the most important ones. The quality, safety and, side effects, knowledge about herbs can be determined while standardizing each herbal formulation. The knowledge about the herbs and herbal medicines by the villagers and tribes is unknown to scientists. Scientists are well-known about the standardization process unknown by rural peoples and tribes. Hence the present study is carried out to identify the bioactive phytochemical constituents present in Amukkara Choornam by spectroscopic analysis. Standardization is a tool in the quality control process must for rural health care. Siddha medicines are our traditional medicines, which have lesser side effects. Since, it is a plant-based product.

Background: The Hepatitis C virus (HCV) is a dangerous infectious illness that has long been a concern for public health on a worldwide scale. According to calculation, over 170 million individuals are afflicted, with the highest infection rates in Africa and Asia. HCV is a major concern worldwide and eliminating it in its early phase to avoid liver cirrhosis and HCC has long been the aim for studies. To date, there is no verified vaccine available in the market and current approved therapy (standard of care). Siddha formulas have managed pathogenic infections for ages. Siddha practice strengthens the host's immunity and resilience to pathogens. Materials and Methods: The main aim of the present investigation is to screen the anti-viral potential of the siddha formulation Manjal noi Kudineer against Hepatitis C viral -RNA dependent RNA polymerase (RdRp) using in-silico docking technique. Results: Result of the study clearly emphasise that the lead cucurbitacin E ranks top with greatest binding free energy -8.96 kcal/mol, followed by Piperidine (-7.71), Curcumin (-7.45), Piperic acid (-6.77), Beta-Humulene (-6.40), Cinnamic acid (-5.89), Oleic acid (-5.59) and Piperine (-5.32). It was also evident that the therapeutics such as oleic acid and Cucurbitacin E revealing potential binding affinity in targeting the activation loop (Leu474, His475, Ser476 and Tyr477) of the viral polymerase enzyme. Followed by which other leads including Limonene, Beta-Pinene, Cinnamic acid, Anethole, Phyllanthin, Piperic acid, Curcumin and Piperine ranked second by offering prominent interactions with the residual bioactive amino acids (His475, Ser476 and Tyr477). Conclusion: From the data's of the present in-silico screening, it was concluded that the phytochemicals in the siddha formulation Manjal noi Kudineer display strong anti-viral property by blocking the target enzyme target enzyme (Hepatitis C viral polymerase) and thereby considered as a novel drug of choice for the clinical management of hepatitis C viral infection.

<p>Severe acute respiratory syndrome coronavirus (SARS-CoV-2) that cause COVID-19 becomes a global threat and spread its pandemicity across the boundaries. Recent demography issued by WHO forecasting the severity of disease prevalence in more than 200 countries resulted in 16,96,588 laboratory confirmed cases excluding 1,05,952 deaths as on 12 April 2020. Countries like USA (4,92,881), Italy (1,52,271), Spain (1,61,852), Germany (1,20,479) are struggling hard to flatten their epidemic curve of COVID-19. Dynamic strategies are of utmost important in order to manage the crucial spread of SARS-CoV-2. Drug of herbal origin may offer reliable therapeutic opportunity in controlling widespread transmission. It was evident from the scientific outcomes that SARS‐CoV-2 gains access in to the host cell through angiotensin-converting enzyme 2 (ACE2) receptors. Hence drugs that reveals potential binding affinity with core amino acid of ACE-2 may expected to interfere the host-viral interaction. In our present investigation 28 lead molecules from well documented medicinal herbs were subjected to molecular docking analysis targeting ACE2 receptor and their potential of impeding host-viral interface were evaluated. Results of computational analysis signifies that out of 28 ligands nearly 11 bioactive lead molecules exhibit potential binding affinity of about 100% with the target amino acid residue (31 Lys and 353 Lys)<br></p>
