Experimental studies on transformation of the variola virus into the vaccinia virus

Abstract: The transformation of variola virus into vaccinia virus by means of passage through animal systems has been a subject of sei6ntific controversy for more than 100 years. The discussions centered around the question whether the vaeeinia virus represents 1) a separate member of the pox viruses which occur naturally; 2) a result of transformation of the variola virus, or 3) --an equally conceivable possibility, a transformed cowpox virus. The experimental research of the past and of recent time shows three stages.… Show more

“…The live-attenuated Ducapox vaccine (batch number DPV0816, manufacture date August 2016, expiration date August 2017) contains 10 6.5 TCID 50 /mL of strain CaPV298-2, which was isolated in the United Arab Emirates (UAE) and attenuated by 120 serial passages into Vero cells [ 9 ]. The vaccine, originally developed by Onderstepoort Biological Products [ 10 , 11 ] and produced by Highveld Biologicals, Republic of South Africa (RSA), is currently manufactured by the Central Veterinary Research Laboratory (CVRL, Dubai, UAE). A total volume of 11 mL of sterile, distilled water has been used for the reconstitution.…”

“…Claims that smallpox vaccine was simply attenuated smallpox virus [9] are unsound. Vaccines were developed from cowpox in circumstances where contamination with smallpox virus could not have occurred, and all modern controlled attempts to derive vaccine from smallpox virus have failed [2,19]. There is no evidence that smallpox virus used for variolation was becoming consistently attenuated before Jenner started work with cowpox, and it is clear that his use of animal mlaterial marked a radical new departure [2].…”

“…For this reason, the origin of vaccinia, the basis of the smallpox LAV, is lost to history. Prior to the eradication of smallpox, attempts to recreate a vaccinia-like LAV through the passage of the variola virus major or minor in embryonated eggs, tissue culture, and mice, were unsuccessful [124]. Additionally, even when methods of LAV passage are described in detail, as in the case of YFV-17D [32], recreation of the vaccine is not always possible.…”
