Experimental Studies on the Significance of Endotoxin in the Pathogenesis of Brucellosis

Abstract: The participation of an endotoxin in the pathogenesis of brucellosis has received scant recognition although clinical observations have suggested that a Brucella toxin might be responsible for some of the manifestations in the acute phases of the disease. This has been especially obvious in patients whose illness has been due to Brucella melitensis. In general, brucellosis caused by Br. melitensis is a more severe disease than that resulting from Brucella abortus (1). It has also been observed in experimentall… Show more

“…melitensis was used for most of the experiments. The method of preparing this type of endotoxin has been previously described (13). Washed, heat-killed E. coli organisms grown on "Albimi" Brucella agar medium and diluted in an equal volume of sterile saline were employed in many experiments.…”

“…The beneficial effect of hydrocortisone or ACTH in ameliorating or preventing the toxicity of brucella endotoxin in humans conforms with the protective action of the adrenocorticosteroids against endotoxin lethality in mice (10,20). It also suggests that the beneficial action of therapy of acute brucellosis with ACTH (21) or steroids (22) may result in part through a suppression of endotoxin activity.…”

“…The properties of brucella endotoxin have been extensively studied in the experimental animal (6)(7)(8) and have been found to be quite similar to those of endotoxins from other organisms (9). Of particular interest are the observations (3,10) that endotoxins extracted from various strains of the three brucella species have identical toxic and immunologic actions, regardless of the capacity of the individual strain to establish infection. In man, the injection of endotoxins from various enteric microorganisms causes fever, headache, chilliness, myalgia, emesis, leukopenia, alterations in carbohydrate metabolism, and vasomotor disturbances which may culminate in shock if sufficient endotoxin is given (11,12).…”

“…In eleven patients the picture resembled an undulant type of fever; the interval between the two waves varying from 2 to 9 months and in two the illness during the second wave was more severe than during the first. It is suggested that these examples might support the view that sensitization of the body to the brucella endotoxin is of importance in the pathogenesis of brucellosis (Spink & Anderson, 1954;Abernethy & Spink, 1958).…”
