Experimental Osteoarthritis Models in Mice

Abstract: Osteoarthritis (OA) is a slowly progressing, degenerative disorder of synovial joints culminating in the irreversible destruction of articular cartilage and subchondral bone. It affects almost everyone over the age of 65 and influences life quality of affected individuals with enormous costs to the health care system. Current therapeutic strategies seek to ameliorate pain and increase mobility; however, to date none of them halts disease progression or regenerates damaged cartilage or bone. Thus, there is an u… Show more

“…First, medial radial displacement (MRD) of the medial meniscus is considered a signature feature of knee OA. The cause of the protrusion is unknown, but MRD may play a hitherto unappreciated role in the pathogenesis of OA due to its close relationship with the medial collateral ligament 14. Bevers et al 15 were the first to study the interobserver reliability reporting a moderate κ for two observers.…”

“…The proteoglycan content of the articular cartilage was graded on Safranin-O-stained sections using OARSI histology scoring system as previously reported [12]. Moreover, the histological analyses of cartilage destruction were done as previously reported [13].…”

“…Nine rats were randomly sacrificed at two days, four days, three weeks, four weeks, and six weeks subsequent to the surgery. The OA model was generated via surgery, according to a previous publication [25]. In short, the ligament cruciatum genu anterius or posterius, the meniscus medialis, and the ligamentum mediale of the posterior rat limbs were eliminated in order to bring about articular deformity, which led to the enhancement of damage to the articular face, as well as deterioration of joint cartilage.…”
