Expanded expression of Sonic Hedgehog inAstyanaxcavefish:multiple consequences on forebrain development and evolution

Abstract: Ventral midline Sonic Hedgehog (Shh) signalling is crucial for growth and patterning of the embryonic forebrain. Here, we report how enhanced Shh midline signalling affects the evolution of telencephalic and diencephalic neuronal patterning in the blind cavefish Astyanax mexicanus, a teleost fish closely related to zebrafish. A comparison between cave-and surface-dwelling forms of Astyanax shows that cavefish display larger Shh expression in all anterior midline domains throughout development. This does not af… Show more

“…Such comparisons are difficult to make in mammals, because deep evolutionary distances separate lineages and considerable size variation obscures brain adaptations (11,16,30). Our conclusion extends those from studies of cavefishes and birds (31,32), and suggests that evolution might capture any point along the continuum of patterning and neurogenesis as neural systems diversify.…”

“…Alternatively, this result could be an artifact of lab rearing. For example, the large adults may have learned that they can better acquire food by responding to olfactory (Menuet et al, 2007) or auditory cues, rather than oscillatory flow when fed in aquaria. Large Pachón cavefish also showed no VAB (Fig.…”

“…The anterior expression domain of the morphogen sonic hedgehog (Shh) at the embryonic ventral midline is expanded throughout cavefish forebrain development (Menuet et al, 2007;Yamamoto et al, 2004). The secondary loss of eyes is the most spectacular indirect consequence of this increased Shh signalling (Yamamoto et al, 2004).…”

“…However, expanded Shh also causes significant and specific modifications in the ventral telencephalon and hypothalamus. In fact, Shh-dependent variations in proliferative activity and in expression of Nkx and LIMhomeodomain transcription factors induce variations in specification and migrations of specific neuronal populations (Menuet et al, 2007). Besides these specific changes, the global early patterning of the forebrain, including the retina, is normal in cavefish.…”
