Exosomal Noncoding RNAs and Tumor Drug Resistance

Abstract: ◥Tumor drug resistance is a major challenge in the treatment of cancer. Noncoding RNAs (ncRNA) play a role in the progression of drug resistance. Recent studies have indicated that exosomes, with their in vitro and in vivo compatibility, are the best natural carrier of ncRNA, and their transport of ncRNA into cells could regulate drug resistance. Exosomal ncRNA impact drug resistance through participation in drug efflux, regulation of signaling pathways, and modification of the tumor microenvironment. In this … Show more

“…Thus, EVs are appreciated as essential mediators of cell-to-cell communication. In the field of cancer biology, EVs have been found to play crucial roles in cancer metastasis [ 189 ] and drug resistance [ 190 , 191 , 192 , 193 , 194 , 195 ]. In oral cancer, EVs also mediate drug resistance by transferring their cargoes to recipient cells [ 196 , 197 , 198 , 199 , 200 , 201 , 202 ].…”

“…Another effective strategy for exosome-targeted therapy is to inhibit the uptake of exosomes with recipient cells. Heparan sulfate proteoglycans (HSPGs) are internalizing receptors of cancer-derived exosomes which could significantly attenuate exosomes uptake [129]. In addition, Reg3β, a lectin binding to the surface of exosomes, was released by normal pancreatic tissue surrounding tumors.…”

“…Even worse, treatment can promote metastasis and cancer lethality (D'Alterio et al, 2020 ; Pienta et al, 2020a , b ). There are many viewpoints/assumptions to explain the mechanisms of drug resistance: multiple drug resistance, cancer stem cells, cellular adaptation and collaboration, epigenetic regulation, cancer heterogenicity, persistent cancer cells, exosomal non-coding RNAs, acquired immune-resistance, and tumor micro-environments including tumor-stromal cross-talk (Heng et al, 2010 ; Holohan et al, 2013 ; Keating et al, 2013 ; Niero et al, 2014 ; Restifo et al, 2016 ; Cho and Kim, 2020 ; Guo et al, 2020 ; Shen et al, 2020 ; Swayden et al, 2020 ; Bhattacharya et al, 2021 ). Among them, new gene mutations and, in particular, selection of preexisting variants by drug treatment are the most popular mechanisms of cancer drug resistance (Khong and Restifo, 2002 ; Aktipis et al, 2011 ; Heng, 2015 , 2019 ).…”
