Exosomal lncRNA GAS5 regulates the apoptosis of macrophages and vascular endothelial cells in atherosclerosis

Abstract: Atherosclerosis is universally recognized as a chronic lipid-induced inflammation of the vessel wall. Oxidized low density lipoprotein (oxLDL) drives the onset of atherogenesis involving macrophages and endothelial cells (ECs). Our earlier work showed that expression of long noncoding RNA-growth arrest-specific 5 (lncRNA GAS5) was significantly increased in the plaque of atherosclerosis collected from patients and animal models. In this study, we found that knockdown of lncRNA GAS5 reduced the apoptosis of THP… Show more

“…They also participate in the pathology of atherosclerosis such as EC dysfunction, cholesterol accumulation, and inflammation (9). For example, lncRNAgrowth arrest-specific 5 (GAS5) expression was significantly increased in the atherosclerotic plaque and exaggerated inflammation in ox-LDL-treated macrophages (10,11). LncRNA-DYNLRB2-2 was upregulated in ox-LDL-induced THP-1 macrophage-derived foam cells and could promote cholesterol efflux and inhibit inflammation (12).…”

“…Several lncRNAs, like lncRNA NRON [3], lncRNA MALAT1 [4], and lncRNA GAS5 [5], have been recognized as angiogenesis regulators by controlling of endothelial cells proliferation, tube formation, and migration. Highly upregulated liver cancer (HULC) is a 500 nt lncRNA.…”

“…Additionally, GAS5 suppressed the miR‐221 expression as a sponge, which might cause plaque destabilization . After oxLDL treatment, lncRNA GAS5 was abundant in THP‐1‐derived exosomes and enhanced vascular endothelial cell apoptosis by sucking up the exosomes …”
