Exopolysaccharide Produced by Lactobacillus paracasei IJH-SONE68 Prevents and Improves the Picryl Chloride-Induced Contact Dermatitis

Abstract: Allergic disease is one of the most important and common health problems worldwide. We have previously demonstrated that a fig leaf-derived lactic acid bacterium Lactobacillus (Lb.) paracasei IJH-SONE68 produces a novel exopolysaccharide (EPS). Furthermore, we have shown that the EPS inhibits the catalytic activity of hyaluronidase (EC 3.2.1.36) promoting inflammatory reactions. To evaluate the anti-allergy and anti-inflammatory effects of the EPS, in the present study, we employed the picryl-chloride-induced … Show more

“…In an in vivo model of AD on BALB/cAJcl mice, the oral administration of an exopolysaccharide (EPS) produced by Lactobacillus paracasei reduced ear swelling, produced a repression of ear interleukin-4 (T helper (Th) 2 cytokine) mRNA and decreased serum immunoglobulin E levels. These results suggest that Lactobacillus paracasei -derived EPS inhibits the catalytic activity of hyaluronidase promoting inflammatory reactions and is useful for improving type I and type IV allergies, including AD [ 69 ].…”

“…In fact, one of the EPSs produced by Lb. paracasei IJH-SONE68 has been reported to display prevention effect in picryl chloride-induced contact dermatitis model mice [ 25 ]. Furthermore, a clinical trial in subjects with allergic conditions using the IJH-SONE68 strain is in progress (UMIN Clinical Trials Registry, trial No.…”

“…Prior to the molecular mass and constitution sugar analyses, the GI-containing fraction was further purified based on the EPS purification procedure described previously [ 25 ]: the concentrated GI fraction prepared from the culture supernatant of BM53-1 was mixed with the same volume of acetone and incubated overnight at 4 °C. The precipitate was collected and dissolved into a 50 mM Tris-HCl buffer (pH 8.0) and treated with nuclease and proteinase K. The GI collected by ethanol precipitation was dissolved into distilled water and applied to the anion exchange resin (TOYOPEARL DEAE-650M, Tosoh Bioscience, Tokyo, Japan).…”

“…We have already isolated over 1000 LAB strains from many plant sources and stored them in a plant-derived LAB library. We have found that some of the stored LAB strains may be useful for preventive medicine, such as immune modulation, reduction of obesity, and anti-allergy [17][18][19][20]. Interestingly, the yogurt manufactured with Lactobacillus (Lb.)…”
