Exogenous Nitric Oxide and Endogenous Glucose-Stimulated β-Cell Nitric Oxide Augment Insulin Release

Abstract: The role nitric oxide (NO) plays in physiological insulin secretion has been controversial. Here we present evidence that exogenous NO stimulates insulin secretion, and that endogenous NO production occurs and is involved in the regulation of insulin release. Radioimmunoassay measurement of insulin release and a dynamic assay of exocytosis using the dye FM1-43 demonstrated that three different NO donors-hydroxylamine (HA), sodium nitroprusside, and 3-morpholinosydnonimine (SIN-1)-each stimulated a marked incre… Show more

“…More recent studies suggest that cyclic GMP could mediate the effects of low concentrations of nitric oxide in augmenting glucose-induced Ca 2+ oscillations and insulin secretion in rat-isolated beta cells [48,49,50]. 8-bromoguanosine-cyclic GMP was found to augment glucose-induced Ca 2+ oscillations in mouse islets and to inhibit K ATP channel activity in mouse-intact beta cells [51].…”

“…All membrane potential measurements were done at 37°C. Images were captured and analyzed using the Merlin software (Olympus America, Life Sciences Resources, Melville, NY) as described elsewhere (13,24). Exposure time was 0.2 s, and images were acquired at ϳ0.2 Hz.…”

“…22,34 Enhancing several pathways involved in insulin gene expression, exogenous NO has been shown to stimulate insulin secretion. When cultured on PA-YK-NO nanomatrices, glucose-stimulated insulin secretion responses (22 mM) for both 3 and 7 days of cultivation were either better or at the same level as the control, indicating that NO in low concentrations may have a beneficial effect on the functionality of MIN-6 cells.…”

“…The lack of NO donors that can release NO in a controlled, sustained manner can lead to results that cannot be reliably interpreted. 22 Some studies suggest a detrimental role, where NO mediates the inflammatory reaction in response to engrafted islet cells. [23][24][25][26][27] This has been evidenced by elevated expression of interleukin-1β and inducible nitric oxide synthase in regions of pancreatic islet allotransplantation.…”
